Artificial Culture of Cordyceps Militaris and the Studies of Its Nucleosides

[Abstract]

The artificial culture of Cordyceps militaris as well as the extraction, isolation, determination, pharmacological activity of their metabolite梟ucleosides were reported in this paper. The optimal ram medium and suitable inoculum time were obtained using growth and spores yield as marker. The optimal liquid medium and solid medium were also confirmed using fermentation rates and dry weight of stroma as marker. Meanwhile, the optimal culture conditions for liquid culture were established.Adenosine and cordycepin were purified from cultured stroma. The studies showed that there were similar nucleosides in natural and cultured Cordyceps militaris. These nucleosides were identified by means of HPCE. The content of cordycepin in cultured stroma and mycelium were 5.04 and 10.85 folds higher than those of natural stroma respectively. The content of cordycepin in solid cultured whiteand yellow mycelium were 57.67mg/g and 85.16mg/g respectively.The pharmacological tests showed that there was the inhibition action of nucleosides in cultured stroma on the hepatocarcinoma H22 cell . It was also showed that nucleosides may improve the healthy balance and survival time of the mices with hepatocarcinoma.
